本物種為經由其與已知角鴞不同的叫聲，於2016年所發現的物種。[6]研究人員根據其形態、羽毛顏色和花紋、叫聲和分子證據於2022年時描述。[5]根據當地人的說法，本物種最早可能在1928年時就已有所紀錄。[5]屬名Otus來自拉丁語的同名詞，指短耳鴞，此詞源自希臘語的ὦτος（羅馬化：ōtos），意為「有耳朵的貓頭鷹」。[7]種小名bikegila則來自當地一位原為鸚鵡獵人，現為自然嚮導的當地居民Ceciliano do Bom Jesus，其外號為Bikegila，研究人員根據其證言發現了這種角鴞。[5]
普林西比角鴞的分佈範圍僅侷限於普林西比島上僅34.4平方公里大的聖多美奧博自然公園（英语：Parque Natural Obô de São Tomé）的原生森林內，且該地有一部分區域正有開發為小型發電水壩的計畫。[4][11]其生存環境就如同當地的另一特有種普林西比鸫（英语：Príncipe thrush）一樣倍受入侵種白腹長尾猴的威脅。[1][12]目前其族群預測僅有約1000—1500隻左右，因此在《國際自然保護聯盟瀕危物種紅色名錄》中這個物種被列為極危物種。[1]
